摘要
根据GenBank上已发表的Hoxc9基因序列和本实验室前期构建绒山羊cDNA文库中Hoxc9基因部分序列设计引物。利用PCR技术从内蒙古绒山羊血液基因组DNA中扩增了Hoxc9基因,目的基因片段纯化后连接到PMD19-T载体,将鉴定的重组质粒进行测序,测序结果通过与GenBank中序列比对分析,确定扩增序列为Hoxc9基因,将该基因DNA序列提交至GenBank,登录号为DQ873298,DNA序列长为3308bp,cDNA序列长783bp,编码260个氨基酸。在此基础上利用生物信息软件对该序列结构特征进行分析。
According to the Hoxc9 genes sequences announced from GenBank and constructed cDNA library from cashmere goat, primers were designed. The Hoxc9 gene of Inner Mongolian cashmere goat were amplified by PCR. The PCR products were purified and then cloned into PMD19 -T vector, combinant plasmids were sequenced. By blasting with database of GenBank, it was confirmed that PCR products were Hoxc9 gene. Its length of DNA and eDNA was 3308bp and 783bp respectively, which encoded 260 amino acid. ( GeneBank Accession : DQ873298). Furthermore, character of gene sequence were analysised using bioinformation software.
