摘要
目的研究PIWIL2在肝门部胆管癌组织和细胞株中的表达及其与临床病理特征和根治性切除术预后的关系。方法采用免疫组织化学方法检测41例肝门部胆管癌和10例正常胆管组织中PIWIL2的表达;蛋白印迹和细胞免疫荧光技术检测人胆管癌细胞系QBC939和人正常胆管上皮细胞HIBEpic株中PIWIL2的表达。采用Kaplan—Meier法对胆管癌根治术后患者进行生存期的单因素及多因素分析。结果PIWIL2在肝门部胆管癌和QBC939细胞中的表达明显高于对照组织和HIBEpie细胞(P〈0.05);PIWIL2在低、中分化胆管腺癌的表达明显高于高分化腺癌(P〈0.05)。单因素生存分析显示根治术后PIWIL2高表达的生存期较短(P〈0.05)。多因素分析显示PIWIL2的表达水平是影响胆管癌根治术预后的独立危险因素(P〈0.05)。结论PIWIL2在肝门部胆管癌组织和细胞系中表达上调,其表达水平与肿瘤分期及分化程度相关,是影响肝门部胆管癌根治切除预后的独立危险因素。
Objective To explore the relationship between PIWIL2 and clinicopathological characteristics and prognosis after radical resection by detecting the expression of PIWIL2 in hilar cholangiocarcinoma tissues and in cell lines. Methods The expression of PIWIL2 was detected by immunohistochemistry in 41 hilar cholangioearcinomas and 10 control tissues, Western-blot and immunocytofluorescent were used to investigate the PIWIL2 expression in eholangiocarcinoma cell lines QBC939 and bile duct epithelial cell lines HIBEpic. Univariate and multivariate analysis of survival was performed using the Kaplan-Meier curve in hilar cholangiocarcinoma patients who had underwent radical resection. Result The expression of PIWIL2 in hilar cholangiocarcinoma tissues and QBC939 cells were significantly higher than the control tissues and HIBEpic ceils ( P 〈 0.05 ). The expression of PIWIL2 in poor and moderate differentiated cholangiocareinoma was significantly higher than the well differentiated group ( P 〈 0. 05 ). Univariate analysis showed that high expression of PIWIL2 was associated with short-survival time after radical resection ( P 〈 0. 05 ). Multivariate analysis showed that the expression of PIWIL2 was an independent prognostic factor after radical resection of hilar eholangioeareinoma ( P 〈 0.05 ). Conclusions The expression of PIWIL2 was associated with TNM stage and differentiation; PIWIL2 was an independent factor for prognosis after radical resection of hilar cholangioearcinoma.
