摘要 目的探讨TGF-β1基因修饰的肝干细胞对大鼠移植肝脏的免疫耐受的影响。方法采用Kamade的"二袖套法"行SD大鼠对Wistar大鼠的异种原位肝脏移植动物模型,根据处理方式的不同将动物分为对照组(生理盐水和地塞米松)、干细胞组(肝干细胞和地塞米松)和实验组(TGF-β1基因修饰的肝干细胞和地塞米松)。记录各组动物的手术基本情况,观察受鼠的存活时间,相比术后24 h后移植肝组织的肝功能指标冬氨酸转氨酶(AST)、总胆红素(TBIL)、丙氨酸转氨酶(ALT)和白蛋白(ALB),比较各组动物肝脏的病理。结果与对照组和干细胞组相比,实验组动物存活时间明显延长,肝功能好转。实验组AST、TBIL、ALT和ALB与其他两组相比,差异具有统计学意义(P<0.05)。在肝脏病理中,实验组的免疫排斥反应明显降低。结论 TGF-β1基因修饰的肝干细胞对移植肝脏具有保护作用,在急性免疫排斥方面具有明显的抑制作用。 Objective To explore the influence of TGF-β1 modified liver stem cells in immune tolerance of liver transplantation in rats. Methods The models of SD to Wistar rats liver transplantation were set up. The animals were randomly divided into control group, cell stem group and experimental group. Survival time and indicators of liver function were analyzed among three groups. The pathological changes of liver in three groups were compared. Results In experimental group, survival time was prolonged. The indicators of liver function, AST, TBIL, ALT and ALB were statistically different among three groups (P 〈 0.05 ). In pathological changes, the immune rejection was decreased signifi- cantly in experimental group. Conclusion TGF-β1 modified liver stem cells may have protecting role in the implanted liver and decrease the acute immune rejection.
