摘要
报道了波氏假丝酵母(Candida boidinii)启动子的克隆,将克隆的启动子与 Neo基因重组构建了 含Neor基因的表达载体YepNP181,转化波氏假丝酵母,实现了Neor基因在波氏假丝酵母中的表达,使波氏 假丝酵母转化子能在含G418的抗性培养基上生长从而建立了Neor基因──G418选择系统,为外源基因在 波氏假丝酵母中表达创造了条件.
Candida boidinii is one of the methylotrophic yeast with good development and application perspective hut very little study has been done as a gene expression system. The C. boidinii promoter was cloned and the ex-pression vector YepNP181 was constructed by recombining the cloned promoter and Neor gene onto an E. coli -yeast shuttle vector. YEpNP181 was then transformed into C. boidinii and Neor gene on YEpNP181 expressed. The transformants grow on YPD plate containing 300 μg/mL C418 and Neor-G418 selection system was thus established, which facilitates the foreign gene expression in C. boidinii.
