大鼠脑出血后血肿周围脑组织PPARα的表达变化

Expressions of PPARα mRNA and protein in brain tissues around intracerebral hematomas in rats

摘要 目的观察大鼠脑出血后血肿周围组织过氧化物酶体增殖物激活受体α(PPARα)的表达变化。方法取成年雄性Wistar大鼠48只,采用自体血注入法制备脑出血模型。术后6、12、24、48、72、120 h断头取脑,应用荧光定量PCR和蛋白印迹法检测血肿周围及对侧脑组织PPARαmRNA和蛋白的表达。结果与血肿对侧脑组织相比,血肿周围脑组织PPARαmRNA和蛋白表达在出血后6 h显著升高并达高峰(P<0.05),随后降低,至48 h与血肿对侧表达量无明显差异(P>0.05);然后再次升高,120h的表达量与6 h无统计学差异(P>0.05)。结论 PPARα在大鼠脑出血后血肿周围脑组织动态表达变化提示其可能参与脑出血后继发性损伤过程。 Objective To explore the expressions of peroxisome proliferator activated receptor a (PPARot) mRNA and protein in the brain tissues around the hematoma at different times after the intracerebral hemorrhage (ICH). Methods ICH model was made by the intracerebral injection of autologous blood in 48 male Wistar rats, which were randomly divided into six groups of 8 animals each, i.e. the groups of 6, 12, 24, 48, 72 and 120 hours after ICH. The expressions of PPARct mRNA and protein in the cerebral tissues contralateral to and around the hematomas in all the groups were determined respectively by quantitative RT-PCR and Western Blot method. Results The levels of PPARot mRNA and protein expressions were significantly higher in the cerebral tissues around the hematomas 6, 12, 24, 72 and 120 hours after the hemorrhage than those in the cerebral tissues contralateral to the hematomas (P〈0.05), but there was no significant difference in levels of PPARct mRNA and protein expressions 48 hours after the hemorrhage between the cerebral tissues contralatered to and ones around the hematomas (P〉0.05). The levels of PPARct mRNA and protein expressions were significantly higher 6 and 120 hours after the hematoma than those 12, 24, 48, and 72 hours after the hemorrhage in the cerebral tissues around the hematoma (P〈0.05), but there were insignificant differences in the levels of PPAR ct mRNA and protein expressions in the cerebral tissues around the hematomas between 6 and 120 hours after the hemorrhage (P〉0.05). Conclusions That the levels of PPARot mRNA and protein expressions in the brain tissue around hematomas increase rapidly after hemorrhage, then decrease gradually to the normal level and thereafter they increase again suggested that PPARa may be involved in the process of the injury secondary to ICH in rats.
