摘要
免疫捕获粪便悬液中的HAV ,RT -PCR扩增目的片段 ,将其连接到载体pUC18中 ,测序并对序列进行分析和比较 .HAV野毒株W在核苷酸 3 0 2 4~ 3 191与HM 175 /wt序列相同 ,表明W株为ⅠB亚型 ;该株5’NTR的核苷酸 2 9~ 2 5 8与其它国际毒株有很大的不同 ,有碱基的替换、缺失和插入 ,其中插入的 15个碱基是其它毒株所没有的 .
HAV particles were captured from clinical fecal suspension by immunoaffinity.The viral fragment synthesized by RT-PCR was recombined into vector pUC18,and analyzed after sequencing.The sequence of HAV wild type W is the same as HM175/wt at nt3?024—3?191,it showed that strain W is sub type ⅠB.However,substitutions,deletion and insertions in 5'NTR at nt58—294 of HAV strain W were detected as compared to other strains isolated from different countries.The inserted 15 bases were not found in any other HAV strains.
