大白菜热激转录因子基因家族鉴定及表达分析 被引量:11

摘要 本文以大白菜基因组数据为基础,利用生物信息学方法对大白菜热激转录因子(heatshocktranscriptionfactors,HSFs)进行了序列特征分析。结果表明,大白菜至少含有30个HSF基因成员,蛋白质序列长度在239~487氨基酸之间;序列多重比对发现,大白菜HSF基因具有高度保守的DBD结构域和广泛的保守基序;染色体定位揭示了除2个基因分布在Scaffold上,其余28个基因在大白菜的10条染色体上呈不均匀分布,Chr03上分布最多;此外,大白菜和拟南芥HSF基因的系统发育关系比较分析表明,这些HSF基因家族成员可以分为A、B、C三类,A类和B类可进一步分为8个和4个亚组,并且存在8对直系同源基因和4对旁系同源基因,表明HSF基因家族存在于芸薹属和拟南芥属分化之前。RT—qPCR分析表明,高温胁迫显著了诱导了大部分大白菜HSF转录因子基因的表达,表明这些基因广泛地参与了大白菜苗期叶片对高温热激反应。 In this paper, genome-wide identification and analysis of heat shock transcription factors (HSFs) in Chinese cabbage were performed with bioinformatics methods. The results showed that Chinese cabbage contains at least 30 HSF genes. The length of protein sequence ranged from 239 to 487aa. The multiple sequence alignment analysis showed that BraHsfs have highly conserved domain of DBD and a wide range of conservative motif. Except for two genes, which were located on the Scaffold, all the other genes are found to be distributed unevenly in the 10 chromosomes with the maximum in chr03. In addition, based on phylogenetic relationship between BraHsfs and AtHsfs in Arabidopsis thaliana, BraHsfs members can be divided into A, B and C class. A and B classes could be further divided into eight and four subclasses. Eight pairs of orthologous genes and four pairs of paralogous genes were revealed, indicating that HSF gene families have existed before the differentiation of Brassica and Arabidopsis. RT-qPCR analysis found that high temperature induced different expression levels of BraHSF in Chinese cabbage, suggesting that these genes were involved in Chinese cabbage response to high temperature.
