不同剂量银杏叶提取物对红藻氨酸致痫大鼠神经肽Y蛋白表达的影响 被引量:3

Influence of different doses of ginkgo biloba extract on neuropeptide Y protein expression in kainic acid induced epileptic rats

摘要 目的:观察不同剂量银杏叶提取物(GBE)对癫痫大鼠的抗癫痫效应及其对神经肽Y(NPY)蛋白表达的影响。方法:采用侧脑室注射红藻氨酸建立大鼠癫痫动物模型。将90只SD大鼠随机分为假手术组、GBE对照组、癫痫模型组、小剂量GBE组、大剂量GBE组,各组再分为6、12、24 h亚组(n=6)。应用HE染色观察海马组织神经元的细胞形态变化,用免疫组化方法检测海马组织中NPY蛋白的表达。结果:GBE对照组、癫痫模型组、小剂量GBE组、大剂量GBE组NPY蛋白的表达均呈上升趋势(F=17.335、60.901、109.587、118.669,P均<0.001)。造模后第6、12、24 h,5组NPY蛋白表达水平比较,差异有统计学意义(F=30.859、126.500、87.401,P均<0.05);与假手术组比较,癫痫模型组、GBE组NPY蛋白表达均升高(P均<0.05);与癫痫模型组比较,GBE组NPY蛋白表达升高(P均<0.05)。结论:2种剂量GBE对致痫大鼠均有一定的脑保护作用。 Aim:To investigate the effects of different doses of ginkgo biloba extract ( GBE) on epileptic rats and the relationship with the expression of neuropeptide Y ( NPY) .Methods:An epileptic animal model was established in rats by injection of kainic acid .A total of 90 SD rats were randomly divided into sham operation group , GBE control group , epilep-tic model group , small dose of GBE group and large dose of GBE group .Each group was further divided into three sub-groups according to observation time .Morphologic changes of the hippocampal neurons were observed by HE staining , while the expression of NPY in hippocampal tissue was measured by immunohistochemical technique .Results:The expres-sion of NPY increased with observation time in GBE control group , epileptic model group and GBE groups (F=17.335, 60.901,109.587,118.669,P〈0.001).NPY expressions of five groups had significant differences at 3 time points(F=30.859,126.500,87.401,P〈0.05).Compared with sham operation group,the expression of NPY in epileptic model group and GBE groups were higher , and that was the highest in GBE groups (P〈0.05).Conclusion:Both large and small doses of GBE have protective effects on brain tissue of epileptic rats .
