摘要
以茄子为研究对象,初步建立起茄子的ISSR和SSR分子标记技术体系,包括DNA提取、退火温度、循环次数和引物筛选等。结果表明,ISSR分子标记体系中,从49条引物中筛选出3个条带清晰的引物对6个品种进行扩增,共检测出26条带,其中多态性带21条,多态性比例为80.77%;SSR分子标记体系中,从4对引物中筛选出3个条带清晰的引物对6个品种进行扩增,共检测出17条带,其中多态性带5条,多态性比例为29.41%,2种方法均能把6个茄子品种区分开来,本实验为茄子品种鉴定和新品种选育提供理论依据。
The study aimed to set up the ISSR and SSR reaction system of eggplant, include the genomic DNA extraction, annealing temperatare, cycle time, primers screening. The results showes in ISSR reaction system, 3 primers were screened from 49 ISSR primers totally to amplify the eggplant genome DNAs of 6 materials effectively,26 sites were amplified out totally, including 21 polymorphism sites with polymorphism rate of 80.77% ;while in SSR reaction system,3 primers were screened from 4 SSR primers totally to amplify the eggplant genome DNAs of 6 materials effectively, 17 sites were amplified out totally,including 5 polymorphism sites with polymorphism rate of 29.41%. All of the 6 varieties could be differentiated from each other based on two methods. The results provide theoretical to the seed purity identification and new varieties breeding choose of the eggplant.
