摘要
[目的]初步建立了涂层毛细管胶束电动色谱快速测定三聚氰胺含量的简便方法.[方法]以40 mmol/L磷酸二氢钠和20mmol/L十二烷基硫酸钠(SDS)为分离缓冲溶液,以石英毛细管为分离柱,于234 nm波长处检测,采用峰高外标法定量.通过对分离缓冲液的组分、石英毛细管涂层、pH及电压对三聚氰胺分析的影响进行研究.[结果]该方法检出限0.48 mg/L,线性范围为10 ~ 500mg/L,线性相关系数0.998 5,三聚氰胺出峰时间小于0.8 min.[结论]通过改进样品处理和试验操作方法,提高回收率,可望满足乳制品中三聚氰胺的定性、定量测定.
[Objective] A preliminary,simple,and rapid micellar electrokinetic chromatography (MEKC) method was established for determination of melamine using coated capillary.[Method] The chromatographic separation was carried out in a coated capillary with a separation buffer composed of 40 mmol/L disodium hydrogen phosphate and 20 mmol/L sodium dodecyl sulfate (SDS).Melamine was detected at 234 nm and quantified by peak-height external standard method.The effects of components of buffer,trimethylchlorosilane coating,pH and applied voltage were investigated.[Result] The detection limits of the method were 0.48 mg/L.The linear range was from 10 to 500 mg/L with a correlation coefficient of 0.998 5.Melamine peak appeared less than 0.8 min.[Conclusion] Through perfecting sample treatment and experiment operation method,the recovery rate was improved,which can satisfy the qualitative and quantitative determination of melamine in dairy products.
