微粒在动脉粥样硬化发生发展中的作用 被引量:9

摘要 微粒(MP)是活化或者凋亡的细胞以出芽方式产生的微小囊状双层膜结构,没有细胞核,直径约0.4~1μm。MP的形成机制尚不完全清楚。离体试验提示,细胞激活或者凋亡时细胞膜双层磷脂结构的非对称性受到破坏,使磷脂酰丝氨酸(PS)暴露在外层,随即细胞以出芽的方式形成MP。循环中的MP来源于不同的细胞,如红细胞、粒细胞、单核细胞、淋巴细胞、血小板和内皮细胞(EC),以血小板来源的MP(PMP)为最多,占循环中MP的70%~90%。其他部位也存在MP,比如动脉粥样硬化斑块,其形成的MP主要集中在斑块局部,不进入循环系统。研究发现MP生成的现象是细胞的正常生理功能,健康人群中可以检测到MP,但细胞在不同的生理、病理状态下产生的MP,在磷脂构成、表面标记物、内部的蛋白组成和基因信息上都存在差异。近年来,MP的生理、病理作用被逐渐认识,其过度产生可见于心血管疾病、肿瘤、感染及正常或者病理妊娠等多种状态,广泛参与细胞黏附、凋亡、免疫应答、血管重塑和生成、止血和血栓形成等过程。特别是MP与动脉粥样硬化斑块的形成、进展及血栓事件的关联性备受重视,本文就该部分内容进行文献综述。
