摘要
目的:研究创伤后应激障碍(PTSD)样大鼠伏隔核神经元内多巴胺D2受体表达变化.方法:用连续单一刺激(SPS)方法刺激大鼠制作PTSD模型,取SPS后12h、1d、7d大鼠脑组织;非刺激的脑组织为对照组,分别用免疫组织化学和免疫印迹进行各组伏隔核中神经元多巴胺D2受体表达的观察.结果:免疫组织化学和免疫印迹结果显示,经SPS处理后,伏隔核神经元多巴胺D2受体表达于刺激后12h低于正常组,1d时可见高于正常,7d时仍处于较高水平.结论:大鼠经SPS处理后,伏隔核内多巴胺D2受体表达先降低后升高,提示该受体表达与PTSD血浆糖皮质激素变化有关,进而促进PTSD的精神症状的产生.
Objective: To study the change of the expression of dopamine D2 receptor (DRD2) in the nucleus accumbens in rats with posttraumatic stress disorder (PTSD). Methods: The single-prolonged stress (SPS) method was used to stimulus the rat to establish a rat PTSD model. The brain of rats was obtained at 12 h, 1 d and 7 d after SPS and normal brain of rats as a control group. Then the expressions of DRD2 in the nucleus accumbens in each group were observed by immunohistochemical techniques and Western blotting. Results: The results of immunohistochemistry and Western blotting showed: after the rats were handled by SPS, the expression of DRD2 in 12 h group in nucleus accumbens was lower than that of the normal, but the expression of DRD2 in 1 d group was higher than that of normal, and that of the 7 d group was still at a higher level. Conclusion: After handled by SPS, the expression of DRD2 in the nucleus accumbens was declined firstly and then increased. It suggests that the expression of DRD2 relates to changes of plasma glucocorticoid in PTSD, thus contributing to the production of the psychiatric symptoms of PTSD.
