大鼠大脑中动脉栓塞对脊髓前角运动神经元乙酰胆碱酯酶的影响

摘要 目的:探讨大鼠大脑中动脉栓塞(MCAO)对脊髓前角运动神经元外侧核群乙酰胆碱酯酶(AChE)的影响.方法:20只雄性SD大鼠随机分为4组,均采用线栓法建立一侧大鼠MCAO致脑缺血模型,术后大鼠分别存活1、2、4、8周,取脊髓颈膨大.采用亚铁氰化铜法显示AChE,Image-Pro Plus 6.0图像分析软件对脊髓前角运动神经元外侧核群AChE反应阳性区域的累积光密度(IOD)进行定量分析.结果:MCAO 1周组实验侧脊髓前角运动神经元外侧核群AChE的IOD值高于对照侧,MCAO 2周组开始低于对照侧,且病程越长此变化更为明显.结论:大鼠MCAO可导致脊髓前角运动神经元外侧核群AChE的含量发生改变. Objective: To investigate the effects of middle cerebral artery occlusion (MCAO) on AChE in the anterior horn motor neurons later nucleus in rats. Methods: 20 male SD rats were randomly divided into 4 groups, the cerebral ischemia on one side was established by MCAO rat model on the same side, and the rats were allowed to survive for 1 week, 2 weeks, 4 weeks and 8 weeks after operation. The cervical enlargement was taken and displayed by Karnovsky-Roots. Image-Pro Plus 6.0 image analysis system was used to analyze the integrated optical density (IOD) of AChE positive region. Results: The AChE IOD of the anterior horn motor neurons of lateral nucleus in 1 week group on the experimental side was higher than that on the control side. The AChE IOD in 2 weeks group on the experimental side was lower than that on the control side. The changes were more obvious with the extension of the course of disease. Conclusion: MCAO in rats can lead to the diversification of AChE in the anterior horn motor neurons of lateral nucleus.
