摘要
目的:总结自噬与肿瘤相关的研究现状,以及在肿瘤诊断、治疗及预后中的作用。方法:应用PubMed及CNKI全文数据库检索系统,以"自噬(Autophagy)、自噬小体(Autophagosome)、细胞凋亡(Apoptosis)、肿瘤(Cancer)"为关键词,检索2009-01-2013-03的相关文献,共421篇英文文献和54篇中文文献。纳入标准:1)自噬的调控机制及相关因子;2)自噬对机体细胞凋亡的影响;3)肿瘤的发病、治疗及预后与自噬的相关性。根据纳入标准符合分析的文献20篇。结果:自噬是真核细胞通过降解细胞质内自身结构来维持自我平衡的代谢过程,其在维持蛋白代谢平衡及细胞内环境的稳定、清除废物、结构重建以及细胞生长发育以及凋亡中起重要的作用。结论:自噬过程受一系列复杂信号分子的调控。调控机制的失调与肿瘤的病理生理过程存在密切联系。自噬因为对细胞凋亡有重要影响而与肿瘤的发病、治疗和预后密切相关。
OBJECTIVE: To sum up the related research association between cancer with autophagy and the role of autophagy in the diagnosis and treatment of cancer. METHODS: Totally 421 English articles and 54 Chinese articles were searched by "autophagy, Autophagosome Apoptosis, cancer" as keywords in PubMed and CNKI databases from Jan. 2009 to Mar. 2013. Totally 20 papers were selected and analyzed according to the inclusion criteria as follows: the mechanisms and related factors of autophagy regulation; Functions of autophagy in apoptosis; The relationship between autophagy and the incidence, treatment and prognosis of cancer. RERULTS: Autophagy, which occurs in all eukaryotic cells, is a process of self-balancing through degradating of cytoplasmic structure. It plays an important role in maintaining the bal- ance of protein metabolism and intracellular stability of the environment, clearing waste, remodeling, cell growth,devel- opment and apoptosis. CONCLUST1ONS: The regulation of this process is affected by a complex series of signaling mole- cules. The effectiveness of regulatory mechanisms has closing contact with the process of cancer. Thus it is widely recog- nized that autophagy has a close relationship with the incidence, treatment, prognosis of cancer as a result of the influence of autophagy on apoptosis.
