连接蛋白43mRNA在妊娠期糖尿病胎盘组织中的表达及意义 被引量:1

摘要 目的探讨连接蛋白43(Connexin43,Cx43)mRNA在妊娠糖尿病(gestational diabetes mellitus,GDM)胎盘组织中的表达。方法收取GDM未治疗组18例,GDM治疗组22例,正常妊娠组12例,HE染色观察各组胎盘组织标本细胞滋养细胞、合体滋养细胞等的变化。应用RT-PCR方法检测Cx43 mRNA在三组胎盘组织中的表达。结果组织学观察显示GDM未治疗组的胎盘组织中细胞滋养细胞、合体滋养细胞明显多于GDM治疗组及正常对照组。三组胎盘组织中均有Cx43 mRNA的表达,Cx43 mRNA在GDM未治疗组较正常妊娠组和GDM治疗组降低,具有统计学意义(P<0.05)。结论 Cx43 mRNA在GDM胎盘中的低表达是引起胎盘结构和功能变化的重要因素,在GDM的发病过程中起重要作用。 Objective To investigate the expression of connexin43 (Cx43) mRNA in placentas from pregnant women with gestational diabetes mellitus (GDM). Methods Placental samples were obtained from 18 untreated cases of GDM, 22 treated cases of GDM and 12 normal controls.Observe the quantity change of placental trophoblastic ceils, syncytial knots in different groups, the quantity of Cx43 mRNA was analyzed by RT-PCR. Results Histologically, the quantity of syncytiotrophoblastic and cytotrophoblastic ceils in the placenta of untreated cases of GDM increased obviously com- pared with treated cases of GDM and normal controls. Cx43 mRNA expressed in all groups were analyzed by RT-PCR. Compared with treated cases of GDM and normal controls, the ratio of untreated cases of GDM decreased significantly (P〈0.05). Conclusion The low expression of Cx43 mRNA resultes in changes of placentas from pregnant women with GDM, which may play important roles in GDM.
