摘要
目的观察HO-1对肺气肿大鼠模型肺组织ICAM-1和MIP-2表达的影响。方法复制肺气肿大鼠模型,并用Hemin干预,收集BALF行细胞计数并分类检查;采用免疫组织化法检测大鼠支气管肺组织HO-1、ICAM-1表达,ELISA方法检测肺组织匀浆中MIP-2的含量。结果与模型组比较,干预组肺组织中HO-1表达显著增加(P<0.05)。肺气肿组大鼠模型中不仅ICAM-1在支气管肺组织的表达较干预组显著增加(P<0.05),而且BALF中的白细胞计数和中性粒细胞计数均与ICAM-1表达呈显著正相关(r=0.85、0.84,P均<0.01);模型组肺组织匀浆中MIP-2含量较干预组明显增加,而且BALF中的白细胞计数和中性粒细胞计数均与MIP-2含量呈显著正相关(r=0.89、0.89,P均<0.01)。结论 ICAM-1和MIP-2在肺气肿中明显增加,HO-1可降低ICAM-1、MIP-2的表达,这可能是HO-1在肺气肿中抗炎反应的作用机制之一。
Objective To observe the effect of HO-1 on expressions of ICAM-1 and MIP-2 in lung tissue of rats with emphysema. Methods Established emphysema rat models and using Hemin to interven. BALF were collected to count the total and differential cell counts. HO-1 and ICAM-1 expressions in pulmonary tissues of rat were detected by im- munohistochemistry. The content of MIP-2 in lung tissues was detected by ELISA. Results Compared to the emphyse- ma group, the expression of HO-1 in control group were significantly positive (P〈0.05). The expression of ICAM-1 in lung of the emphysema group was significantly higher than those in control group (P〈0.05) and was positively correlated with the leukocyte numbers and neutrophil in BALF in the model group (r=0.85, 0.84 respectively, P all 〈0.01). The content of MIP-2 in lung tissues of the emphysema were significantly higher than those in control group(P〈O.05). In the model group, the leukocyte numbers and neutrophil in BALF were positively correlated with the levels of MIP-2 in lung tissues (r=0.89, 0.89 respectively, P all 〈0.01). Conclusion HO-1 might significantly alleviate emphysema by inhibiting the expressions of ICAM-1 and MIP-2.
