腓肠肌斜刺联合环孢素A对软组织钝挫伤模型兔IGF-1表达的影响 被引量:5

Effect of CsA and Oblique Needling the Gastrocnemius on the Expression of Insulin-like Growth Factor 1 in Rabbit Models With Soft Tissue Injury

摘要 目的观察腓肠肌斜刺联合环孢素A(CsA)对软组织钝挫伤修复的影响及其作用机制。方法 30只新西兰大白兔随机分为空白组、模型组、CsA组、针刺组、针刺合并CsA组,每组6只。采用"重物自由落体打击法"连续20次撞击造成急性软组织钝挫伤模型。针刺组造模后24h后采用阿是穴斜刺干预,CsA组于造模前3天开始腹腔注射CsA 9.25 mg/(kg·d),针刺合并CsA组采用针刺合并CsA干预,模型组和空白组不干预。于损伤后72h采用Real-time PCR、蛋白免疫印迹法(Western blot)方法检测各组腓肠肌胰岛素样生长因子-1(IGF-1)mRNA及其蛋白表达量。结果与空白组相比,其余各组IGF-1 mRNA表达均显著降低(P<0.05或P<0.01),模型组、CsA组与针刺组IGF-1蛋白表达显著降低(P<0.05或P<0.01)。与模型组相比,针刺组和针刺合并CsA组IGF-1 mRNA与蛋白表达显著升高(P<0.05);与CsA组相比,针刺组与针刺合并CsA组IGF-1 mRNA及其蛋白表达均显著升高(P<0.05)。结论软组织钝挫伤可使兔骨骼肌IGF-1 mRNA及其蛋白表达降低,腓肠肌斜刺联合CsA可上调急性期骨骼肌损伤局部IGF-1mRNA及其蛋白表达,促进软组织修复。 Objective To observe the effect of cyclosporine A (CsA) and oblique needling into the gastrocnemi us on soft tissue injury (STI) and its mechanism. Methods Thirty New Zealand white rabbits were randomized in to blank group, model group, CsA group, needling group and needling combined with CsA group, with 6 in each. The acute STI models were established by heavy objects in free fall for 20 consecutive shocks. The needling group was given oblique insertion of the needles into Ashi points, CsA group was given intraperitoneal injection of 9.25 rag/ (kg . d) CsA and needling combined with CsA group was given both needling and CsA 24 h after modeling. There was no intervention in the model group and blank group. The expressions of insulin-like growth factor 1 ( IGF-1 ) mRNA and its protein in gastrocnemius were detected with Real-time PCR and Western blot 72 hours after injury. Results Comparing with the blank group, the expression of IGF-1 mRNA was significantly decreased in other groups (P 〈0. 05 or P 〈0.01 ) and the expression of its protein was significantly decreased in the model group, CsA group and needling group (P 〈0. 05 or P 〈0.01 ). Comparing with the model group, the expressions of IGF-1 mRNA and its protein were significantly increased in the needling group and needling combined with CsA group (P 〈 0. 05 ). Comparing with the CsA group, the expressions of IGF-1 mRNA and its protein were significantly increased in the needling group and needling combined with CsA group (P 〈 0. 05 ). Conclusion STI can decrease the expressions of IGF-1 mRNA and its protein. The combo therapy of CsA and inserting the needles obliquely into the gastrocnemius can up-regulate the expressions of IGF-1 mRNA and its protein in partially injured skeletal muscles, and improve the soft tissue repairment.
