摘要 目的:研究姜黄素对肺腺癌A549细胞增殖率、凋亡和PI3K/Akt/Bcl-2信号通路的影响。方法:采用细胞计数法绘制肺腺癌A549细胞生长曲线,MTT法测定姜黄素对肺腺癌A549细胞抑制率。将不同浓度姜黄素作用于肺腺癌A549细胞24h后,显微镜下观察细胞形态;TUNEL法检测细胞凋亡率;免疫组化法检测PI3K/Akt/Bcl-2蛋白表达。结果:姜黄素能够抑制肺腺癌A549细胞增殖,且呈一定剂量和时间依赖性;随着药物浓度上升细胞凋亡率明显增高(P<0.05);PI3K、Akt和Bcl-2蛋白表达明显下降(P<0.05)。结论:姜黄素对肺腺癌A549细胞增殖具有抑制作用,其作用可能是诱导细胞凋亡及抑制PI3K/Akt/Bcl-2信号通路来实现的。 Objective:To explore the effect of curcumin on PI3K/Akt/Bcl -2 Pathway Signaling of lung adenocarcinoma cell A549. Methods : Cell counting was used to plot the cell growth curves of A549 cells. The inhibition rate of A549 cell in different concentrations of curcumin were measured by MTI assay. After the cells were treated with curcumin, cellular morphology was observed under micro scope. The apoptosis rate was detected by TUNEL. PI3K/Akt/Bcl - 2 proteins were detected by two - step immunohistochemical staining. Results : Curcumin could restrain the proliferation of A549 cells and inhibitory effect was dose - and - time - dependent. The rate of apoptosis increased significantly with the increasing concentrations of the drug, The expression of PI3K/Akt/Bcl - 2 proteins were decreased significantly( P 〈 0.05 ). Conclusion : Curcumin exerts inhibitory effects on proliferation of A549 cells possibly through indu cing cell cycle apoptosis and inhibition of PI3K/Akt/Bcl - 2 pathway signaling.
