摘要
目的探讨芷辛方的镇痛作用及对骨癌痛大鼠模型中脊髓胶质细胞蛋白(GFAP、CD11b)表达的影响。方法将SD大鼠,随机分为空白组(10只)、骨癌痛模型组(10只)、西药对照组(10只)、芷辛高剂量组(10只)、芷辛正常剂量组(10只)、芷辛低剂量组(10只)、中药对照组(10只)。造模14天后开始灌胃,每日一次,连续7 d。对各组的大鼠进行行为学检测、X光片检测及HE染色。第21天行为学测试后,取脊髓L4-L5部位,分别应用免疫组化及RT-PCR检验星形胶质细胞的标志性蛋白GFAP的变化以及小胶质细胞的标志性蛋白CD11b表达变化。结果骨癌痛模型造模成功。免疫组化检测骨癌痛模型组的髓内星形胶质细胞的标记蛋白GFAP,随时间的推移,呈现明显增生肥大。PCR检测CD11b,骨癌痛模型组明显高于芷辛正常剂量和芷辛高剂量(P<0.01)。结论芷辛方具有镇痛作用,抑制脊髓水平胶质细胞的增殖活化可能是其机制之一。
Objective To explore the analgesic effect of Zhixin Formula (ZF) and its effects on spinal glial fibrillary acidic pro- tein (GFAP) in rats with bone cancer pain. Methods SD rats were randomly divided into seven groups, 10 in control group, 10in model group, 10 in western medicine group, 10 in ZF high dose group, 10 in ZF normal dose group and 10 in ZF low dose group. On dl4, rats were with administrated by garage for seven days, once a day. Later, behavioral examination, X -ray and HE staining were applied to evaluate modeling. On d21, MS typical protein expression including GFAP and CD11 b from spinal IA - L5 was detected by immunohistochemistry and RT - PCR. Results Models were successfully constructed. Immunohistochemis- try showed that GFAP expression in model group proliferated and hypertrophied significantly. RT- PCT showed that CD1 lb ex- pression in model group was significantly higher than that in ZF normal dose group and ZF high dose group (P 〈 0.01 ). Conclu- sion ZF has analgesic effect, which is probably related to its inhibition of gliocyte proliferation and hypertrophy.
