摘要
目的研究白花蛇舌草对CEM细胞抑制作用及机制。方法以不同浓度的白花蛇舌草水提物作用CEM细胞24,48,72h后,通过MTT比色试验,检测其对CEM细胞的抑制率;用浓度分别为166μg/ml、33.2μg/ml、6.64μg/ml的白花蛇舌草水提物作用于CEM细胞24,48,72h后,通过逆转录PCR(RT-PCR)检测P53基因的表达,探索白花蛇舌草抑制CEM细胞的可能机制。结果白花蛇舌草水提物能够抑制CEM细胞的生长;白花蛇舌草能促进CEM细胞的P53基因表达,且随用药浓度的升高及作用时间的延长,其抑制作用逐渐增强、P53基因表达也逐渐增强。结论白花蛇舌草水提物能抑制白血病CEM细胞的生长,其作用强度与时间、浓度呈正相关,其抑制机制可能与上调P53基因的表达有较大关联。
Objective To research the possible inhibition mechanism of Hedyotis diffusa Willd on CEM cell. Methods With dif- ferent concentration of Hedyotis diffusae aqueous extract affected on CEM cells last 24,48,72h, using MTT colorimetric assay to detect CEM cell inhibition ratio; with concentrations of 166μg/ml,33.2μg/ml,6.64μg/ml Hedyotis diffusae aqueous extract af- fected on CEM cells last 24,48,72h, using reverse transcription PCR( RT - PCR) to detect the expression of P53 gene, to ex- plore the possible inhibition mechanism of Hedyotis diffusae on CEM cells. Results Hedyotis diffusae Willd aqueous extract can inhibit the growth of CEM cells ; the expression of P53 gene in CEM cells can be inhibit by Hedyotis diffusae Wilid, and with the increase of drug concentration and action time prolonged , the inhibitory effect was enhanced. Conclusion Hedyotis diffusae willd aqueous extract can inhibit Leukemia CEM cells growth. The effect of intensity has positive related with the use of time and the concentration of drug. The inhibition mechanism of cells growth has greater correlation with the up - regulation of P53 expression.
