摘要
目的探究黄芪中皂苷与多糖及两者配伍对脾淋巴细胞免疫活性的影响。方法制备小鼠脾淋巴细胞悬液,依次进行正常培养和ConA(刀豆蛋白)诱导培养,采用MTT(溴化四唑蓝)微量比色法检测淋巴细胞OD值,分别考察黄芪皂苷、多糖单用及二者配伍对淋巴细胞体外增殖的影响,并于培养结束前取各孔培养液上清,检测NO含量。结果对正常培养的小鼠脾淋巴细胞,黄芪皂苷在高浓度1,0.2 mg/ml对细胞增殖具有明显抑制作用,低浓度为促增殖作用,而单用多糖均促进细胞增殖,不同比例配伍时以皂苷0.2、多糖1.6 mg/ml促增殖作用最强,此配伍浓度下NO达最大值,皂苷1.6、多糖0.2 mg/ml配伍抑制作用最强,NO含量亦随之降低,而等比配伍时高浓度抑制,低浓度促进;对ConA诱导培养的小鼠脾淋巴细胞,黄芪皂苷、多糖均表现为抑制作用,等比配伍时均显著抑制细胞增殖以及NO的释放,且有明显浓度依赖性。结论黄芪皂苷、多糖不同的配伍比例、配伍浓度其免疫调节活性会发生相应变化,对细胞增殖的影响与NO释放量相关,不同比例配伍时其综合效应随浓度增加一方发生改变,等比配伍时则表现为高浓度拮抗,低浓度协同或单纯相加。
Objective To explore the immune activity variation dealt by astrageloside (AS), astragalus polysaccharide (APS) and their compatibility to normal training and ConA - activated cultured mouse lymphocyte. Methods Preparing spleen lympho- cytes in mice suspension continue to culture 48 hours, to detect NO content and the influence of lymphocyte proliferation normal and activated by ConA with MTT of AS , APS and their compatibility in vitro. Results To the normal cultured spleen lymphocytes, AS could inhibit lymphocyte proliferation under high concentrations, promote proliferation under low concentration, and APS could promote lymphocyte proliferation. The ratio of AS and APS is 0.2: 1.6 showed the strongest effect on promoting proliferation and NO content contrary to the ratio of AS and APS is 1.6: 0.2. The effect on cell proliferation of Astragalus composition geometric compatibility showed obvious inhibitory effect under high concentration contrary to low concentration. To the lymphocytes activated by ConA, different concentrations of AS or APS were characterized by inhibition, and geometric compatibility all inhibit the growth of proliferation and NO content in a dose dependent manner. Conclusion The combined effect on immune regulating of Astragalus component compatibility change with the concentration and ratio in compatibility. It is found that different ratios compatibility show similar tendency in lymphocyte proliferation with high concentrations and equal ratio compatibility are antagonistic effect under high concentration contrary to low concentrations, which are in correlation to NO content.
