正交实验法优选四味清口含片中挥发油的β-环糊精包合工艺 被引量:5

Orthogonal experimental design for optimization of inclusion process for volatile oil in Siweiqing buccal tablets with β-cyclodextrin

摘要 目的采用饱和水溶液法研究β-环糊精(β-CD)包合四味清口含片中广陈皮、桂心、细辛挥发油的最佳工艺。方法采用正交实验设计L9(34),以油和β-CD比、搅拌速度、包合温度、包合时间为考察因素,以挥发油利用率和包合物收率进行综合评分为评价指标,优选β-CD包合四味清口含片中挥发油的最佳工艺条件。结果最佳挥发油包合工艺条件为:挥发油与β-CD比为1∶4,搅拌速度为100 r/min,包合温度为50℃,包合时间为3 h。结论该工艺操作简便、稳定、可行,挥发油的利用率和包合物收率都较高,为进一步的工艺开发提供参考。 Objective Based on the saturated water solution preparation method, the inclusion process conditions for volatile oil from Citrus Chacbiensis Hortorum, Cortex Cinnamomi and Asarum heterotropoides Fr. Schmidt var. mandshuricum ( Maxim. ) Kitag. in Siweiqing buccal tablets with β -cyelodextrin (fl - CD) had been optimized. Methods The inclusion process was opti- mized by using the orthogonal experimental design, while the utilization ratio of volatile oil and the yield of inclusion compoundswere adopted as indexes. The influence of the four factors such as the ratio of volatile oil to β- CD, the stirring speed, and inclu- sion temperature and inclusion time was discussed. Results The optimum parameters of preparation conditions for inclusion com- pounds are a ratio of volatile oil to β- CD 1: 4, a stirring speed in 100 r/min, an inclusion temperature at 50 ℃ ,and an inclusion time of 3 h, respectively. Conclusion The inclusion process is simple, stable and feasible for operation, and high in utilization ratio of volatile oils and the yield of inclusion compounds. It is also worthy of further trial in technical development.
