NF-κB在脓毒症大鼠肺组织中的活性变化及炎调方对其活性的影响 被引量:10

摘要 目的探讨炎调方对脓毒症大鼠肺组织核因子-κB(NF-κB)活性及血清高迁移率族蛋白B1(HMGB1)水平变化的影响。方法将雄性SD大鼠随机分为正常组、假手术组、模型组和炎调方组。盲肠结扎穿孔术(CLP)制备脓毒症大鼠模型,于造模后2,8,12,24h处死大鼠,凝胶迁移电泳法(EMSA)检测右肺组织NF-κB活性,双抗体夹心酶联免疫吸附法(ELISA)测定HMGB1水平。结果正常组及假手术组大鼠肺组织NF-κB微弱活化,模型组中,CLP术后2h NF-κB活性开始增强,24h仍有较强的活性;炎调方组与模型组相比,NF-κB活性各时间点均明显减弱。模型组HMGB1水平2h开始升高,炎调方组与模型组相比,HMGB1水平明显降低(P<0.05)。结论炎调方能抑制脓毒症大鼠肺组织NF-κB活性,降低脓毒症大鼠血清HMGB1水平。 Objective To investigate the regulating effect of yantiaofang on the activation of nuclear factor - κB ( NF -κB ) in lung tissue and the serumal level of HMGB1 with sepsis in the rats. Methods Male SD rats were randomly divided into four groups:normal control group, sham operation group, cecal ligation and puncture (CLP) group and Yantiaofang pretreated group. Cecal ligation and puncture (CLP) were performed to make sepsis model. Rats were sacrificed at 2,8,12 and 24h,the level of the HMGB1 was examined with ELSA and right lung tissue was obtained to test the activation of NF -κB with electrophoretic mobility shift assay (EMSA). Results The activation of NF - κB could be detected in lung tissue in very low values in normal control group. In the CLP group and Yantiaofang pretreated group,the activation of NF - κB enhanced at 2h and peaked at 8h. Compared with the CLP group,the activation of NF - κB in Yantiaofang pretreated group attenuated obviously, especially at 2 and 8h. The level of HMGB1 in CLP group increased at 2h and peaked at 8h. Compared with the normal control group, the level of HMGB1 in- creased in CLP group (P 〈 0.01 ). The level of HMGB1 in Yantiaofang pretreated group was much lower than that in CLP group (P 〈 0.01 ) besides 24h group. Conclusion The Yantiaofang can inhibit the activation of NF- κB and down -regulate the level of the HMGB1.
