CYLD的亚细胞定位及其对微管组织的调节

摘要 CYLD基因位于16q12.1,编码一个由956个氨基酸组成的蛋白质,其基因的突变与人类家族性圆柱瘤等肿瘤的发生有着密切的关系.利用从细菌中纯化的CYLD片段免疫小鼠,得到CYLD特异性抗体.通过该抗体对细胞进行免疫荧光显微分析,发现在有丝分裂末期内源的CYLD与微管共定位于中体上.进一步利用绿色荧光蛋白标记的CYLD转染细胞,发现CYLD过表达导致细胞内微管组织的紊乱.这些研究结果提示CYLD作为一种微管结合蛋白,除了已知的调节微管组装的动态性之外,在调节微管的组织方面可能也发挥重要的作用. CYLD(cylindromatosis),agene located on chromosome 16q12.1,encodes a protein consisting of 956 amino acids,and its mutation has been implicated in the development of human familial cylindroma.In this study,a CYLD-specific antibody was prepared by immunization of mice with bacterially purified CYLD fragment.Immunofluorescence microscopic analysis of cells using this antibody revealed that endogenous CYLD colocalized with microtubules at the midbody during telophase.Overexpression of CYLD increased the percentage of cells with disorganized microtubules.These results suggest that CYLD,as a microtubule-binding protein,might play an important role in modulating microtubule organization,in addition to its known role in regulating microtubule assembly dynamics.
