子痫前期患者蜕膜组织中RORγt mRNA和Foxp3 mRNA的表达

The expression levels of RORγt mRNA and Foxp3 mRNA in deciduas of patients with pre- eclampsia

摘要 目的研究子痫前期患者蜕膜组织中Th17和Treg细胞转录因子的表达水平,并探讨其在子痫前期发病机制中的作用。方法选取重度子痫前期患者蜕膜组织32例作为实验组,健康妊娠妇女蜕膜组织32例作为对照组,采用RTPCR方法,检测各组研究对象中RORγt mRNA和Foxp3 mRNA的表达水平。结果 1.子痫前期重度组蜕膜组织中RORγt mRNA水平明显高于对照组(P<0.01),而Foxp3 mRNA水平明显低于对照组(P<0.01)。2.RORγt mRNA/Foxp3mRNA在子痫前期重度组的比值高于对照组(P<0.05)。结论子痫前期患者蜕膜组织中存在明显的Th17/Treg细胞免疫失衡,可能是导致子痫前期发生的重要原因。 Objective: To investigate the expression levels of Th17 cells and Treg cells' s transcriptions factors and to explore their role in pathogenesis in patient's deciduas with pre- eclampsia. Methods: Select preeclampsia( severe) patients decidua 32 cases as the experimental group,32 women with normal pregnancy decidua as a control group,using RT- PCR method to detect the maternal decidua RORγt mRNA and Foxp3 mRNA levels expression. Results: 1. There was significant increase in Th17 cell and RORγt mRNA level in patients deciduas with pre- eclampsia( P 0. 01). Meanwhile,there was significant reduction in Treg cell frequency and Foxp3 mRNA level in patients deciduas with pre- eclampsia( P 0. 01). 2. The ratio of RORγt mRNA / Foxp3 mRNA in deciduas of patients with pre- eclampsia is higher than normal pregnancy group( P 0. 05). Conclusion: The deciduas Th17 / Treg imbalance may plays an important role in pre- eclampsia.
