Expression and clinical significance of c-IAP1 in cervical cancer

摘要 目的:检测凋亡相关基因在宫颈癌中的表达,探讨维吾尔族宫颈癌的发病机制及与临床病理指标的关系。方法:采用实时荧光定量PCR及免疫组化对45例宫颈癌及21例正常宫颈组织中c-IAP1进行检测,并分析其表达水平与相应临床病理指标的关系。结果:凋亡抑制基因c-IAP1在宫颈癌中表达上调,c-IAP1mRNA在宫颈癌中的表达量是正常宫颈组织c-IAP1mRNA表达量的3.39倍。与正常宫颈相比,宫颈癌组织中c-IAP1mRNA在不同临床分期、有无淋巴结、肿瘤直径、肌层浸润均呈现高表达,呈显著正相关(P<0.05),与病理分化程度无关。在蛋白水平,c-IAP1在宫颈癌的不同临床分期、有无淋巴结、肿瘤直径、临床分期、肌层浸润及病理分级中的表达差异均有统计学意义(P<0.05)。结论:宫颈癌组织中c-IAP1的高表达可能在宫颈癌抗凋亡机制中起着重要作用,这对宫颈癌的易感性预测、早期诊断、疾病监测及其分子靶向治疗的开展也具有重要的指导意义。 Objective: To detect the expression of apoptosis - related genes in cervical cancer, explore the relationship between pathogenesis of cervical cancer in women of Uighur nationality and clinicopathological parameters. Methods : Real - time fluorescent quanti- tative PCR and immunohistochemistry were used to detect the expression levels of c - IAP1 in 45 cases with cervical cancer and 21 cases with normal cervix. The relationship between expression level of c - IAP1 and corresponding clinicopathological parameters was analyzed. Re- suits: The expression of c - IAP1 in cervical cancer was upregulated, the expression level of c - IAP1 mRNA in cervical cancer was 3.39 times than that in normal cervix. Compared with normal cervix, the expression levels of c - IAP1 mRNA in cervical cancer with different clin- ical stages, lymph node metastasis or not, different tumor diameter, myometrial invasion were high, there was a significantly positive correla- tion (P 〈 0.05 ), which was not correlated with degree of pathological differentiation. There were statistically significant differences in ex- pression levels of c - IAP1 protein among cervical cancer with different clinical stages, lymph node metastasis or not, different tumor diame- ter, myometrial invasion and pathological grades (P 〈 0.05 ) . Conclusion: High expression of c - IAP1 in cervical cancer may play an im- portant role in antiapoptotic mechanism of cervical cancer, which also has important guiding significance for susceptivity prediction, early di- agnosis, disease monitoring and the development of molecular targeted therapy.
