摘要
为了研究克隆五指山猪Myf5(myogenic factor 5,Myf5)基因,获得基因序列并分析基因结构及相关遗传变异,试验采用PCR扩增五指山猪Myf5基因组序列,通过克隆测序寻找基因中的突变位点。结果表明:在五指山猪Myf5基因组中共发现3处突变位点,分别位于第1外显子(C+121G,C+288G)和第1内含子(C+1 204G),生物信息学分析发现,五指山猪Myf5基因启动子区域存在一些潜在的转录因子结合位点,例如Sp1、GATA-1、C/EBPb、CdxA、SRY、Nkx-2和AML-1a等。
To study and clone the myogenic factor 5 ( Myf5 ) gene of Wuzhishan pig for obtaining the gene sequence, and analyze its gene stroc- ture and related genetic variation. The Myt5 genomic of Wuzhishan pig sequence was amplified by PCR, and then the gene mutation sites were found by cloning and sequencing. The results showed that three mutation sites were detected in the Myf5 genome of Wuzhishan pig, which were located in the first exon ( C + 121 G, C + 288G) and the first intron ( C + 1 204G), respectively. Bioinformatics analysis found that there were some potential transcription factor binding sites in the promoter region of the porcine Myi5 gene of Wuzhishan pig, such as the sites for Spl, GATA-1, C/EBPb, CdxA, SRY, Nkx-2, AML-la and so on.
