毛冬青组培苗壮苗生根与炼苗移栽技术研究 被引量:7

摘要 目的:以前期诱导培养的毛冬青无菌组培苗为试材,进行壮苗生根和炼苗移栽的试验研究。方法:对不同植物生长调节剂浓度配比进行选择,用正交设计法筛选出最适宜的壮苗生根培养基;对开盖时间、水培时间、保留有效叶片数及不同基质进行考察,以筛选出最佳的炼苗移栽条件。结果:适宜的壮苗培养基为MS+6-BA 0.3mg·L-1+NAA 0.5 mg·L-1,适宜的生根培养基为1/2 MS+NAA 0.2 mg·L-1+IBA 0.1 mg·L-1;开盖炼苗和水培练苗最适宜时间为8 d,保留叶片10片以上成活率最高,最佳的炼苗移栽基质为蛭石,成活率达87.50%。结论:6-BA和NAA对壮苗有显著影响,NAA效果强于6-BA,NAA对根的诱导有显著意义;移栽成活的关键是适当地进行炼苗,蛭石良好的保水透气性和保肥能力更适于毛冬青的移栽。 Objective: Rapid propagation in vitro and transplant technique of Ilex pubescenas were studied with aseptic seedlings. Methods: Orthogonal design method was used to investigate the effect of combinations of different plant growth regulartors on strengthening seedling and rooting of Ilex pubescen. The effect of duration of opening vessel lid, the length of hydroponic time, the number of remained leaves and different type of transplanting stromata on survival ratio of seedlings were appraised in order to screen out the best transplant conditions. Results: The results showed that the suitable phytohormonie medium for strengthening seedling was MS +6-BA 0.3 mg-L-~ + NAA 0.5 mg'L-1 , the fitting rooting medium was 1/2 MS + NAA 0. 2 mg. L- 1 + IBA 0. 1 mg. L- 1, The best time of opening vessel lid and the best hydroponic time were both 8 days, keeping more than 10 pieces of leaves could increase the survival rate, the best transplant stromata was vermiculite, whose survival rate was up to 87.50%. Conclusion: The research showed that both 6-BA and NAA had prominent effect on strengthening seedling, and the effect of NAA was greater than that of 6-BA. In addition, NAA revealed a better effect on the induction of rooting. The key to success of transplant was hardening of seedling. Vermiculite seemed to be more suitable for transplant due to its capacity of keeping moisture, gas permeability and fertilizer.
