摘要
目的探讨雌、孕激素对淋球菌(Ng)诱导的人宫颈癌细胞胸腺基质淋巴细胞生成素(TSLP)和转化生长因子β(TGF-β)表达的影响。方法体外培养宫颈癌细胞株Hela,将获得的Hela细胞分为如下5组:对照组、Ng组、Ng+雌激素组、Ng+孕激素组、Ng+雌激素+孕激素组。分别培养24h后应用实时荧光定量PCR检测各组细胞TSLP和TGF-βmRNA表达变化情况,Western blot检测各组细胞TSLP和TGF-β蛋白水平。结果 Hela细胞在淋球菌的刺激下,细胞TSLP和TGF-βmRNA及蛋白表达水平增高(P﹤0.01);雌、孕激素单独或联合作用24h后都可以显著提高Hela细胞中TSLP和TGF-βmRNA表达水平(P﹤0.01)。结论 Hela细胞在淋球菌作用下合成TSLP和TGF-β增加;提示雌、孕可促进淋球菌诱导的TSLP和TGF-β合成。
Objective To investigate the regulatory effects of estrogen and progestin on thymic stromal lymphopoietin (TSLP) and transforming growth factor-β(TGF-β) expression in Hela pretreated with Neisseria gonorrhoeae (Ng). Methods Hela were cultured and randomly divided into five groups: control group, Ng group, Ng + estrogen group, Ng + progestin group and Ng + estrogen + progestin group. Real-time PCR and Western blot was used to examine the mRNA and protein level of TSLP and TGF-β respectively in the Hela cells at 24h after Ng infection. Results The expression ofTSLP ,TGF-β mRNA and protein in Hela ceils at 24h after Ng infection increased significantly than that in the controls( P 〈 O. 01 ). Either estrogen alone, progestin alone or progestin combined with estrogen could significantly increase the expression of TSLP, TGF-β mRNA and pro- tein after 24hours of treatment ( P 〈 0.01 ). Conclusion Nersseria gnnorrhoeae induces the production of TSLP and TGF-13 in Hela cells. Both estrogen and progestin can up-regulate the expression of TSLP and TGF-β gene in Hela cells.
