银丹心脑通软胶囊对大鼠颈动脉血管重构和NF-κB(p65)表达的影响 被引量:4

Effects of Yindan Xinnaotong capsule on vascular remodeling in rats and NF-κB(p65) expression

摘要 目的研究银丹心脑通软胶囊(YDXNT)对球囊损伤所致大鼠颈总动脉重构及NF-κB(p65)表达的影响。方法随机抽取5只大鼠为假手术组,其余大鼠行颈总动脉球囊损伤术造模,存活大鼠随机分为模型组(n=6)、YDXNT低剂量组(n=5)和高剂量组(n=5)。造模后次日开始给药,YDXNT低、高剂量组大鼠灌胃给予YDXNT 0.5、1.0 g·kg-1·d-1;模型组与假手术组大鼠给予生理盐水,每日给药1次,共14 d。HE染色观察损伤侧颈总动脉形态学,计算其内膜面积(NIA)及中膜面积(MA),免疫组化法检测增殖细胞核抗原(PCNA)、NF-κB(p65)的表达。结果与假手术组相比,模型组大鼠颈动脉管腔明显狭窄、内膜增厚;与模型组比较,YDXNT低、高剂量组球囊损伤所致颈动脉管腔狭窄程度显著减轻(P<0.05或P<0.01),颈动脉NIA、NIA/MA明显减小,PCNA、NF-κB(p65)表达下降(P<0.05或P<0.01)。结论 YDXNT可改善球囊损伤所致大鼠颈总动脉重构,可能与其下调NF-κB(p65)表达有关。 AIM To investigate the effects of Yindan Xinnaotong capsule (YDXNT) on vascular remodeling induced by balloon- injury in rats and NF-κB (p65) expression. METHODS Five rats were randomly selected for sham operation. The rat carotid arterial intima-injured model was established with balloon- injured method, and the survival rats were randomly divided into model (n = 6) and YDXNT groups (0.5 or 1.0g·kg^-1·d^-1, n = 5). YDXNT were administered once daily for 14 days in the YDXNT groups, and volume- matched normal saline were administered in the sham and the model groups. After 14 days, the neointimal hyperplasia level and the proliferation of vascular smooth muscle cell (VSMC) were evaluated by calculating the neointimal area (NIA) and the media area (MA). The immunohistochemistry method was used to examine the expression of proliferating cell nuclear antigen (PCNA) , and NF-κB (p65). RESULTS The carotid artery lumen stenosis and neointimal hyperplasia were serious in the model group. Compared with the model group, the carotid artery lumen stenosis induced by balloon injury were remarkably alleviated, the NIA and NIA/MA were decreased and the expressions of PCNA and NF-κB (p65) were attenuated significantly in the YDXNT groups balloon-injury in rats, in which the down-regulate of expression of NF-κB may be involved.
