全细胞色度学分析在鉴别胸水腺癌细胞和间皮细胞中的作用 被引量:3

Effect of whole cell chromatic analysis on differential identification between adenocarcinoma cells and reactive mesothelial cells in pleural effusion

摘要 目的定量观察和分析胸水涂片中腺癌细胞的细胞浆、细胞核和全细胞的色度学特征,筛选对判别ACC和反应性间皮细胞有参考价值的色度学参数。方法胸水离心后取沉淀物涂片、然后进行巴氏染色;在数码显微镜下拍摄图像,用计算机图像分析技术测量其中的764个ACC(44例)和709个RMC(39例)的色度学参数,包括细胞浆、细胞核和全细胞的红、绿、蓝三基色,并对测试结果进行判别分析,比较基于三种色度学参数所建立函数的判别效果。结果与RMC相比,ACC的细胞浆、细胞核和全细胞红、绿、蓝三基色明显减小(P<0.05)。基于以上三种色度学参数所建立函数的判别符合率分别为68.7%、59.4%和74.7%,其中基于全细胞色度学参数所建立的函数判别符合率明显高于基于细胞浆和细胞核色度学参数所建立函数的(P<0.05)。结论在胸水ACC的定量判别分析中,全细胞的色度学参数比细胞浆和细胞核可能更具有参考价值。 Objective The aim of this study was to quantitatively reveal the chromatic characteristics of adenoearcinoma cells (ACC) in pleural effusion, and to find highly specific parameters which could be applied to discriminate ACC from reactive mesothelial cells (RMC) Methods Pleural effusion was centrifuged and the precipitate was taken to make a cell smear, and stained with Digital microscopic images of 764 ACC of 44 cases and 709 RMC of 39 cases were sis,-including chromatic parameters of the three primary colors (red, green, blue Papanicolaou staining. taken for image analy- ) from the cytoplasm, nuclei and whole cells. The results were analyzed by discriminant analysis, and the discriminant coinci- dence of function based on three chromatic parameters was compared. Results Compared with those of RMC, the values of three primary colors of the cytoplasm, nuclei and whole cells in ACC weresignificant- lydecreased (P 〈 0.05). The discriminant coincidencerate of function based on chromatic parameters of the cytoplasm, nuclei and whole cells was 68.7% , 59.4% and 74.7% , respectively, and the coinci- dencerate of function based on chromatic parameters of the whole cell was significantly higher than those based on cytoplasm and nuclei (P 〈 0.05). Conclusions Compared with that of the cytoplasm and nuclei, chromatic parameters of the whole cells might have more valuein quantitative discriminant analysis ofadenocarcinoma cellsinpleural effusion.
