摘要
目的探讨血清基质金属蛋白酶9(MMP-9)水平及其基因C1562T多态性与维族脑梗死患者颈动脉粥样硬化斑块稳定性的关系。方法采用彩色多普勒超声诊断仪对236例维族急性脑梗死患者进行颈动脉超声检测,根据颈动脉粥样硬化斑块稳定性进行分组。采用酶联免疫吸附法检测患者血清MMP-9水平,采用PCR-限制性片段长度多态性技术检测MMP-9基因C1562T的多态性。对结果进行组间比较并分析。结果颈动脉斑块呈低回声或等回声的患者有108例(易损斑块组),颈动脉斑块呈高回声的患者有128例(稳定斑块组)。易损斑块组血清MMP-9水平[(357.83±32.64)ng/ml]显著高于稳定斑块组[(304.47±38.91)ng/ml](P<0.01)。两组MMP-9基因C1562T基因型CC、CT、TT的比例和等位基因C、T的频率比较,差异无统计学意义。结论血清MMP-9水平可能与维族脑梗死患者颈动脉斑块稳定性有关,而MMP-9基因C1562T多态性与维族脑梗死患者颈动脉粥样硬化斑块稳定性无关。
Objective To explore the correlations among the serum levels of matrix metalloproteinase-9 ( MMP- 9) and its gene C1562T polymorphism and the stability of carotid arteriosclerotic plaque in Uygur patients with cerebral infarction. Methods Two hundred and thirty-six Uygur patients with acute cerebral infarction were detected by color Doppler uhrasonography, and were divided according to the carotid atherosclerotic plaque stability. The serum level of MMP-9 was measured by enzyme-linked immunosorbent assay, and the MMP-9 gene C1562T polymorphism was detected by PCR-restriction fragment length polymorphism. The results between the groups were analyzed. Results There were 108 cases with equal-echo or low-echo of carotid plaques (vulnerable plaque group) and 128 cases with high-echo of carotid plaques ( stable plaque group). The serum level of MMP-9 in vulnerable plaque group [ (357.83 ±32.64 ) ng/ml ] was significantly higher than that in stable plaque group [ ( 304.47 ± 38.91 ) ng/ml] (P 〈0.01 ). There were no significant differences of the frequencies of genotype CC, CT, TT and allele C, T of MMP-9 gene C1562T between the vulnerable plaque group and the stable plaque group. Conclusion The serum level of MMP-9 may be correlated with the stability of carotid arteriosclerotic plaque in Uygur patients with cerebral infarction, while the MMP-9 gene C1562T polymorphism may have no correlation with the stability of carotid arteriosclerotic plaque in Uygur patients with cerebral infarction.
