HPLC法测定清热八味散中胆红素的含量

Determination of Bilirubin in Qingrebaweisan by HPLC

目的建立清热八味散中胆红素的含量测定方法。方法采用高效液相色谱法测定清热八味散中胆红素的含量。色谱柱为KromasiL C18柱（5μm，250mm×4．6mm）；流动相为甲醇一三氯甲烷-1％磷酸溶液（70：27：3）；检测波长：450nm；流速：1．0mL·min^-1；柱温：25℃；进样量：10μL。结果胆红素在1．0μg·mL^-1-30μg·mL^-1范围内与峰面积呈良好的线性关系，回归方程为A=39007C＋5936．5，r=0．9994。平均加样回收率为99．89％，RSD为1．86％。结论该方法简便，准确，重复性好，可作为清热八味散中胆红素的含量测定方法。

Objective To establish a method for determination of bilirubin in Qingrebaweisan. Methods Using HPLC to determine the content of bilirubin in Qingrebaweisan. A KromasiL C18 column （250mm × 4.6mm, 5μm） was used. The mobile phase was methanol：chloroform：l% phosphoric acid（70：27：3）, The flow rate was 1.0mLomin1, the detecting wavelength was at 450nm. Results A good linearity of bilirubin was in the range of 1.0-30μg·mL^-1 （r=0.999 4）. The average recovery was 99.89% and RSD was 1.86%. Conclusion This method is simple, accurate and with good reproducibility. It can be used for determination of bilirubin in Qingrebaweisan.
