滇杨多倍体的诱导研究

Study on polyploid induction of Populus yunnanensis Dode

利用组织培养技术,对滇杨进行多倍体诱导,以期获得滇杨多倍体新种质。结果表明:滇杨叶柄预培养5 d后,转接在附加有40%秋水仙素的分化培养基中诱导处理2 d,可获得较好的诱导效果,诱变率高达50.96%。MS+IBA0.10 mg/L+NAA0.10 mg/L为滇杨多倍化组培苗的最佳生根培养方案。对10株性状变异明显的多倍化滇杨植株进行染色体数目观察,发现4株为四倍体,另6株仍为嵌合体。

In order to obtain new polyploid germplasm of Populus yunnanensis Dode,Tissue Culture technique was employed to induce P. yunnanensis polyploid.The results show that after pre-culturing 5 days, the petioles of P.yunnanensis were treated in differential medium supplemented with 40%colchicinefor for 2 days, the best effect could be gotten and the inducing rate was 50.96%;MS＋IBA0.10 mg/L＋NAA0.10 mg/L was the best rooting culture medium formula of polyploid P.yunnanensis;Four tetraploid and six chimera plants were found in ten obviously polyploidy variation by observing the chromosome numbers on ten P. yunnanensis polyploid trees that had obvious phenotypic variances.
