摘要
目的:优化南蛇藤中南蛇藤素的提取工艺,并测定化合物在南蛇藤不同部位中的含量分布。方法:利用箱线图设计(BBD)实验,采用响应曲面法(RSM)优化南蛇藤素的提取工艺,利用毛细管电泳联用技术分析南蛇藤素在南蛇藤不同部位中的分布。结果:南蛇藤素的最佳提取条件为采用80%甲醇,料液比20 ml·g-1,超声提取时间为50 min。南蛇藤素在根、茎、皮、叶中的含量分别为5.13,1.51,0.32,0.34 mg·g-1。结论:结果表明,南蛇藤因用药部位不同,其南蛇藤素含量存在显著差异,响应曲面-毛细管电泳联用技术是一种准确、快速提取并测定物质含量的简便方法。本实验为进一步开发南蛇藤的用药资源提供了理论依据。
OBJECTIVE To optimize the extraction process of celastrot in Celastrus orbiculatus Thunb. ; and to determine its content in different parts. METHODS Using Box-Behnken Design(BBD) and the response surface methodology (RSM) to op timize the extraction process of celastrol, an efficient capillary zone electrophoresis (CZE) analysis method was developed for determination of celastrol in different parts of Celastrus orbiculatus (C. orbiculatus) Thunb. RESULTS The results showed that mixed solvent of methanol and water (80:20,v/v) was the most suitable extractant. The other optimized conditions were solvent to material ratio 20 ml·g^-1 and extraction time 50 min. The contents of celastrol in root,stem, bark and leaf of C. or- biculatus Thunb were 5.13,1.51,0. 32 and 0, 34 mg·g^-1. CONCLUSION There are significant variations for the contents of celastrol in different parts of C. orbiculatus Thunb.. The results indicated that RSM is a handy extraction method with higher yield and lower solvent consumption. The CZE analysis method was validated to have good linearity, precision, reproduction and accuracy. RSM CZE array detection is a fast and appropriate method for determination of celastrol in (2. orticulatus Thunb.. The study provided theoretical foundation for developing medicine resource of C. orticulatus Thunb.
