摘要 将药用野生稻总DNA通过穗茎注射法导入恢复系R225,创制耐荫、耐光氧化的水稻新种质.结果表明,在T0和T1代幼苗期进行全天侯遮光处理15 d后,有90%~75%的秧苗黄化甚至枯萎;在T2和T3代孕穗期和灌浆结实期,通过测定耐光氧化指数,最终筛选到耐荫、耐光氧化的3个导入系;经SSR标记检测,RM219和RM5490在导入系中扩增出了与受体R225不同的3种带型,表明药用野生稻DNA被成功导入受体R225中,并据此推测,标记RM219和RM5490很可能与耐荫或耐光氧化QTL位点紧密连锁. Bred new rice germplasm with photooxidation and shade tolerance by ear-stem injecting the DNA of Oryza officinalis into the restorer R225. About 90%-75% of the To and Tl generation seedlings turned yellow even died after fully shaded day and night for 15 d. Finally, 3 plant lines with photooxidation and shade tolerance were screened by determining the photooxidation-tolerance index in booting and filling stages in T2 and T3 generations. Three types of DNA ladders different from R225 were amplified in transformed lines by SSR makers detecting. The results showed that the DNA of O. officinalis was transformed into R225 successfully. Presumably, RM219 and RM5490 link closely to the QTLs of photooxidation or shade tolerance in flee.
