摘要
以小麦S-腺苷甲硫氨酸脱羧酶(SAMDC)cDNA序列为信息探针,基于高粱EST数据库,通过电子克隆技术获得一个全长为2088 bp的高粱SAMDC基因。采用生物信息学方法对其开放读码框、理化性质、二级结构、结构域等进行分析。结果表明:此contig片段包含一个长达1197 bp的完整开放读码框,编码398个氨基酸,分子量为43 246.0 KD,理论等电点为4.88;二级结构主要构件为无规则卷曲,结构功能域属于S-腺苷甲硫氨酸超家族。同源性分析表明,高粱SAMDC基因编码的蛋白与甘蔗SAMDC基因编码的蛋白同源性达94%。以上研究结果为进一步克隆该基因及功能验证奠定了基础。
The sequence of S-adenosylmethionine decarboxylase (SAMDC) cDNA in wheat was used as a probe,and the homologous in Sorghum was obtained by in silicon cloning.In addition,the bioinformatics characters were analyzed using the methods of bioinfomatics in the following aspects,including general physical and chemical properties,the open reading frame,hydrophobicity and secondary structure.The cDNA length of the gene was 2088 bp long and contained a complete ORF (993 bp) which encoded 398 amino acids.The molecular weight of deduced protein was 43246.0 KD,theoretical isoelectric point of 4.88,the main components of the secondary structure was random coil.The structure and function of the domains belong to the superfamily of S-adenosylmethionine.The amino acid sequence of the gene showed high similarity with sugar cane.All these laid a solid foundation for the gene function analysis.
