摘要
番茄斑萎病毒属(Tospovirus)病毒编码的非结构蛋白NSs,可能在病毒复制、转录、抑制RNA沉默中起重要作用,本文利用生物信息方法对21个Tospovirus病毒成员的NSs序列进行分析,为进一步研究NSs的功能提供参考信息。从GenBank数据库中下载NSs基因序列,用DNAMAN5.0做NSs相似性矩阵分析,发现一些Tospovirus病毒成员之间的NSs氨基酸序列相似性很低,最低仅为7.1%;用ClustalX2.1进行NSs的氨基酸多序列比对分析,发现Tospovirus病毒成员的NSs相对保守区主要位于NSs的C端的位置,高度保守的氨基酸位点共有9个;PROSITE对NSs进行模体预测分析,结果显示Tospovirus病毒NSs共有的蛋白修饰:C蛋白激酶磷酸化、酪蛋白激酶II磷酸化和N-酰基化。此外,不同种的Tospovirus病毒NSs模体也表现出了多样性的特点。预测结果为下一步研究NSs功能奠定了基础。
The nonstructural proteins (NSs) encoded by Tospoviruses were the suppressor of virus,and involved in the replication,transcription of virus.The bioinformatics methods were used to analyze the NSs sequences of 21 Tospoviruses from GenBank to provide further the reference information for studying on function of the NSs.NSs Similarity marx analysis by DNAMANS.0 showed that the low similarities(the lowest:7.1%) existed among NSs amino acid sequence of some Tospoviruses.The result of ClustalX2.0 alignment analysis indicated conserved region of NSs amino acid sequence was located in C-terminal and total of 9 conserved amino acid residue sites were found.The predictive analysis of motifs of NSs proteins using PROSITE demonstrated that common modification of NSs protein include Protein kinase C phosphorylation,Casein kinase Ⅱ phosphorylation and N-myristoylation.And diversities of motifs depended on the species of Tospoviruses.The results would provide the basis for further investigating the function about NSs in future.
