枇杷花化学成分及抗菌抗肿瘤活性研究 被引量:13

摘要 研究枇杷花[Eriobotrya japonica(Thunb.) Lindl.]乙酸乙酯提取物的化学成分及其抗菌、抗肿瘤生物活性,本文运用硅胶、反相ODS、Sephadex LH-20等柱色谱及半制备液相色谱技术对枇杷花乙酸乙酯提取物的化学成分进行分离纯化,根据理化性质及波谱技术鉴定化合物结构,并采用Kirby-Bauer纸片扩散法、MTT法进行体外抗菌、抗肿瘤生物活性的研究.结果表明,分离得到7个化合物,分别鉴定为β-谷甾醇(1)、齐墩果酸(2)、熊果酸(3)、3β,19α-二羟基-4-醛基-乌索-12-烯-28-酸(4)、β-胡萝卜苷(5)、槲皮素3-O-β-D-半乳糖苷(6)、2α-羟基齐墩果酸与2α-羟基乌苏酸(7)(化合物7是由2个化合物组成的混合物),化合物1、4~7首次从枇杷花中分离得到.化合物2、4、5、7具有一定的抗菌活性,人慢性白血病细胞K562对化合物2、3、7敏感,20ug·mL-1浓度下抑制率分别为74.84%、91.96%、74.31%. In order to study the chemical constituents of ethyl acetate extract of flowers of Eriobotrya japonica L.and their antibacterial,antineoplastic activity,in this paper,the chemical constituents of flowers of Eriobotrya japonica L.were isolated and purified by repeated column chromatography on silica gel,ODS chromatographies,Sephadex LH-20 and semi-preparative HPLC,the structures of compounds were identified on the basis of physicochemical and spectroscopic analysis,and the antibacterial and antineoplastic effects of the compounds were measured by Kirby-Bauer and MTT method.The result showed that seven compounds were obtained and identified as β-sitoetero (1),Oleanic acid(2),Ursolic acid(3),3β,19α-dihydroxyursolic-4-aldehyde-12-en-28-acid(4),β-daucosterol (5),quercetin-3-O-α-L-galactoside(6),2α-hydroxyursolic acid and 2α-maslinic acid (7),and compounds 1,4-7 were isolated from this flower for the first time.Compounds 2,4,5,7 exhibited in-vitro antibacterial activity,and compounds 2,3,7(20 μg/mL) exhibited in-vitro antineoplastic activity againat human chronic leukaemia K562 cells with inhibition rstios of 74.84 %,91.96 %,74.31%.
