摘要
利用正交设计试验法研究2,4-D、6-BA、NAA和KT等四个因素及其组合对禾荔花药、叶片及茎段等不同外植体愈伤组织诱导率的影响,以期建立禾荔花药、叶片及茎段愈伤组织诱导技术体系。结果表明:以MS+0.1 g/L Vc+30 mg/L蔗糖+7 mg/L琼脂为基本培养基,花药在添加2.5 mg/L 2,4-D、0.5 mg/L 6-BA和1.0 mg/L NAA的培养基上诱导愈伤组织效果较好;叶片在添加4.0 mg/L 2,4-D、0.5 mg/L 6-BA和1.0 mg/L KT的培养基诱导愈伤组织效果较好;茎段在添加2.5 mg/L 2,4-D、1.0 mg/L 6-BA和0.5 mg/L KT的培养基上诱导愈伤组织效果较好。与叶片及茎段相比,禾荔以花药为外植体诱导产生的愈伤组织质量较高。
The effects of 2,4-D ichlorophenoxy acetic acid,6-BA (6-Benzylaminopurine),NAA (1-naphthlcetic acid) and KT (Forchlorfenuron) as well as their combinations on callus induction of different explants of anther,leaf blade and stem from Litchi chinensis Sonn.were compared based on orthogonal experiment in order to establish callus induction technology system for anther,leaf blade and stem of litchi.The results showed that using MS supplemented with 0.1 g/L Vc,30 g/L sucrose and 7g/L agar as basal medium,the best effect for both induction rate and the quality of the callus induced from the anther was on the basal medium supplemented with 2.5 mg/L 2,4-D 0.5 mg/L6-BA and 1.0 mg/L NAA.The best effect for both induction rate and the quality of the callus induced from the leaf blade was on the basal medium supplemented with 4.0 mg/L 2,4-D,0.5 mg/L6-BA and 1.0 mg/L KT.The best effect for both induction rate and the quality of the callus induced from the stem was on the basal medium supplemented with 2.5 mg/L 2,4-D,1.0 mg/L6-BA and 0.5 mg/L KT for Litchi chinensis Sonn,respectively.Compared with leaf blade and stem,the quality of the callus induced from anther was better.
