透射电镜下电针对慢性应激致抑郁大鼠海马AST结构和功能的影响 被引量:4

Effects of electro-acupuncture on chronic stress which induced depression rats' hippocampal structure and function of AST under TEM

摘要 目的:探讨针刺在修复星形胶质细胞(AST)损伤方面的抗抑郁作用机制。方法:将成年雌性SD大鼠80只,随机分为4组:正常对照组、空白组、电针组和药物组,每组20只;其中正常对照组大鼠进行正常饲养,其余3组采用长期慢性不可预见性温和刺激联合孤养建立抑郁大鼠模型。造模成功后进行行为学检测。空白组经行为学观察造模成功后不予任何处理;电针组经行为学观察造模成功后第2天开始进行电针四关穴治疗,左右两侧穴位交替进行,持续21d。药物组经行为学观察造模成功后第2天开始给予利鲁唑灌胃治疗,连续治疗21d。治疗后进行行为学检测以脱臼法处死大鼠,取大鼠海马组织进行透射电镜观察。结果:行为学糖水偏好实验(SPT)和新环境抑制进食实验(NSFT)检测提示成功建立抑郁大鼠模型接受5周慢性应激后大鼠海马AST超微结构损伤,主要表现为细胞质中粗面内质网明显扩张和线粒体的嵴疏松变。治疗3周后,电针组和药物组AST胞质中粗面内质网扩张和线粒体嵴疏松变均有所减轻,其他细胞器,核糖体、微丝等也较空白组清晰可见。结论:针刺可以修复慢性应激所造成的抑郁大鼠海马星形胶质细胞的损伤,且疗效与利鲁唑相似,这可能是针刺抗抑郁的重要途径和作用靶点之一。 Objective: To explore the antidepressant mechanism of electro-acupuncture in terms of repairing damaged astrocytes. Methods: 80 adult female SD rats were randomly divided into four groups: normal control group, control group, electro-acupuncture group and drug group; the normal control group rats were fed normally, and the remaining three groups were used chronic unpredictable mild stimulation and lone separation to make model. Blank group wasn't given any intervention after successful modeling. Electro-acupuncture group was given acupuncture Siguan point from the second day after successful model according to behavior observation, alternatively operated two sides points. Drug group was intragastricly administered riluzole from the second day after successful model. All treatment sustained 21 days. Rats were sacrificed using dislocation, and hippocampus was carried out to observate under TEM. Results: After 5 weeks of chronic stress, rat hippocampus AST ultrastructure damaged, mainly included the dilated rough endoplasmic reticulum of cytoplasm, and mitochondria crest loose changed. After 3 weeks of treatment, electro-acupuncture and drug group AST cytoplasm rough endoplasmic reticulum and mitochondria are the loose change mitigation, other organelles, ribosomes, filaments, also compared with the control group clearly visible. Conclusion: Electro-acupuncture can fix the depression caused by chronic stress injury in rat hippoeampal astrocytes, and similar efficacy with riluzole, which may be one of the important targets of acupuncture pathways and antidepressant.
