胎膜早破孕妇胎膜组织中基质金属蛋白酶9与金属蛋白酶组织抑制剂2和核因子κB的表达与意义 被引量:3

摘要 目的 探讨胎膜早破患者胎盘胎膜组织中基质金属蛋白酶9(MMP-9)、金属蛋白酶组织抑制剂2(TIMP-2)和核因子κB(NF-κB)表达变化及意义.方法 采用免疫组织化学法,实时定量聚合酶链反应法和免疫印迹法分别检测胎膜早破剖宫产病例45例(胎膜早破组)、正常妊娠择期剖宫产52例(未临产组)、足月自然临产组56例(临产组)孕妇分娩后胎膜组织中MMP-9、TIMP-2和NF-κB的表达水平.结果 实时定量PCR在未临产组,临产组和胎膜早破组中,MMP-9的相对表达量分别是(3.0±1.2)、(10.4±3.1)、(22.1±3.5),TIMP-2的相对表达量分别是(7.1±2.7)、(4.6±2.0)、(3.6±1.8),NF-κBP65在3组的相对表达量分别是(1.1±0.4)、(2.5±0.4)、(3.3±0.4),3组数据比较差异均有统计学意义(P<0.05);免疫印迹显示,未临产、临产和胎膜早破组的MMP-9在胎膜的IOD值分别为(29.5±1.2)、(68.7±3.1)、(167.2±4.8),TIMP-1的IOD值分别是(61.1±4.1),(32.2±2.7),(7.3±3.0),NF-κB P65的IOD值分别是(22.3±3.2)、(52.5±3.6)、(182.7±5.0),3组数据比较差异均有统计学意义(P<0.05).结论 MMP-9在胎膜早破中起到关键性作用,MMP-9/TIMP-2的平衡失调可能间接导致胎膜早破,可能通过NF-κB信号通路发挥调控作用. Objective To discuss the expression and significance of matrix metallo proteinases 9 (MMP-9),tissue inhibitor of matrix metallo proteinase 2 (TIMP-2) and nuclear factor Kappa B (NF-κB) in the fetal membrane of pregnant women with PROM.Methods The expression of MMP-9,TIMP-2 and NF-κB was measured by immunohistochemical method (IHC),realtime polymerase chain reaction(PCR) and Western Blot technique in 45 PROM cases with cesarean section (PROM group),52 normal pregnancy cases with elective cesarean section (not labor onset group) and 56 term labor cases with spontaneous parturient (labor onset group).Results Quantitative PCR showed MMP-9 gene expression in not labor onset group,labor onset group and PROM group was(3.0 ± 1.2),(10.4 ± 3.1),(22.1 ± 3.5) separately.TIMP-2 gene expression in three groups was (7.1 ± 2.7),(4.6 ± 2.0),(3.6 ± 1.8),NF-κB P65 gene expression in the groups was (1.1 ± 0.4),(2.5 ± 0.4),(3.3 ± 0.4),There was statistics difference in groups (P 〈 0.05) about three gene expression.The IOD value of MMP-9 by Western Blot in three groups was (29.5 ± 1.2),(68.7 ± 3.1),(167.2 ± 4.8).The IOD value of TIMP-2 was (61.1 ± 4.1),(32.2 ± 2.7),(7.3 ± 3.0).The IOD value of NF-κB P65 was (22.3 ± 3.2),(52.5 ± 3.6),(182.7 ± 5.0) in not labor onset group,labor onset group and PROM group separately.There was statistics difference in groups(P 〈 0.05) about three protein expression.Conclusions The expression of MMP-9 and TIMP-2 is of negative correlation.The proportion between MMP-9 and TIMP-2 is crucial for PROM and it can take effect through NF-κB signaling.
