摘要
目的:优化玄参的提取工艺。方法:以3种环烯醚萜苷(桃叶珊瑚苷、哈巴苷、哈巴俄苷)和3种苯丙素类(毛蕊花糖苷、安格洛苷C、肉桂酸)的总峰面积为响应值,在单因素试验基础上采用Box-Behnken Design响应面法对甲醇浓度、液料比、超声时间进行优化。结果:最佳提取工艺条件为甲醇浓度88.15%,液料比36.23,超声25.65 min。在此条件下提取总面积预测值为3.12×106,采用改良条件(甲醇浓度88%,液料比36,超声提取26 min)进行验证实验,测得值为3.11×106,实测值与理论值偏差小于1%。结论:响应面法可用于玄参活性成分的提取工艺的优化。
Objective: To optimize the extraction process of Radix Scrophulariae. Methods: A gradientHPLC method was established for the determination of aucubin, harpagide, harpagoside, acetoside, angrosideC and cinnamic acid in Radix Scrophulariae (Xuanshen). The effects of methanol concentration, extractiontime and ratio of solid to liquid on the extraction effciency of three iridoid glycosides and three phenyl-propanoid in Radix Scrophulariae were investigated. The extraction parameters were optimized with Re-sponse Surface Methodology on the basis of single factor experiments using a three-factor and three-levelBox-Behnken center united design. Results: The results showed that the optimum conditions of extractionwere: concentration of methanol 88.15%, ratio of solid to liquid 1:36.23, extraction time 25.65 min. Thepredicted extraction area of six constituents was 3.12×10^6, and the practical extraction area was 3.11×10^6.The RSD between the practical and the predicted values was less than 1%. Conclusion: Box-BehnkenDesign-Response Surface Methodology can efficiently predict the optimum extraction process of Radix Scro-phulariae.
