稳定高表达XAF1基因肺癌细胞株的构建及其对肺癌细胞生物学特性的影响

摘要 目的探讨稳定高表达X连锁的凋亡抑制蛋白相关因子1(XAF1)基因对肺癌细胞生物学特性的影响。方法使用慢病毒作为载体将XAF1基因转染到肺腺癌A549细胞,用灭瘟素试剂(blasticidin)进行筛选,建立稳定高表达XAF1基因细胞株A549,用Western blot法检测细胞生存素蛋白和上皮细胞钙黏蛋白表达的变化。结果成功构建稳定高表达XAF1基因肺癌细胞株A549。A549中XAF1及上皮细胞钙黏蛋白表达水平升高(P<0.05),细胞生存素蛋白表达下降(P<0.05)。结论用慢病毒载体可成功构建稳定表达XAF1基因的肺腺癌细胞株A549。XAF1基因高表达可抑制肺癌细胞的生长和转移。 Objective To investigate the effects of stably expressing X-linked inhibitor of apoptosis(XIAP)-hssociated factor 1(XAF1) on the biological characteristics of lung ad cells. Methods The XAF1 gene was transfected into lung adenocarcinoma A549 cells using lentiviral vector. The A549 cells stably expressing XAF1 were selected by screening with blasticidin. The expressions of survivin and E-cadherin were detected by Western blot. Results A A549 lung cell line was developed, which stably and highly expressed XAF1 with increased expression of E-cadherin(P^0. 05) and decreased expression of survivin (P^0. 05). Conclusion A A549 lung adenocarcinoma cell line has been successfully developed, which stably and highly expresses XAF1. The overexpression of XAF1 can depress the development and metastasis of lung cancer.
