摘要 结冷胶是一种微生物胞外多糖,通过药瓶发酵实验得出结冷胶培养基的最佳培养成分:实验以葡萄糖,麦芽糖,蔗糖,可溶性淀粉,乳糖分别为碳源,其他成分不变,得出最佳碳源为蔗糖,然后通过单因素实验确定最佳用量为3%;以牛肉膏,蛋白胨,酵母膏,豆饼粉,硝酸铵分别为氮源,其他成分不变,得出最佳氮源为豆饼粉,然后通过单因素实验确定最佳用量为0.5%;以MgSO4,KH2PO4,NaCl,KCl,MnCl2,FeSO4分别为无机盐,其他成分不变,得出添加MgSO4和KH2PO4的产量都很高,再通过单因素实验得出最佳用量;最后通过正交实验确定培养基最佳成分:蔗糖为3%,豆饼粉为0.6%,MgSO4为0.06%,KH2PO4为0.14%。 Gellan gum is a microbial extracellular polysaccharide.The optimum fermentation medium of gellan gum was selected by flask shaking fermentation experiments.Glucose,maltose,sucrose,soluble starch and lactose were tested and compared.It was found that the best carbon source was sucrose,and best dosage was 3% which determined by single factor experiment; beef extract peptone,yeast extract,soybean cake powder,and ammonium nitrate were selected as nitrogen source respectively,and after comparison,it was found that the best nitrogen source was soybean cake powder,and the best dosage was 5% determined by single factor experiment; MgSO4,KH2PO4,NaCl,KCl,MnCl2,FeSO4 were chosen as inorganic salt respectively,and MgSO4 and KH2 PO4 both were very good as inorganic salt,the best dosages were also determined by single factor experiment; finally the best composition of the medium through the orthogonal experiment was:sucrose 3%,bean cake powder 0.6%,MgSO4 0.06%,KH2PO4 0.14%
