摘要
目的:研究雌激素受体beta亚型(ERβ)单独活化对CBA/J小鼠Treg细胞的调节作用。方法:应用流式细胞术检测Oil组、E2组、DMSO组、DPN组小鼠胸腺CD4+CD25+Foxp3-GITR+细胞及CD4+CD25+Foxp3+GITR+细胞占CD4+T细胞百分比、小鼠脾脏CD4+CD25+Foxp3+Treg细胞占CD4+T细胞百分比、外周脾表达GITR的Treg细胞百分比及Treg细胞中GITR的平均荧光强度。结果:E2组与Oil组之间、DPN组与DMSO组之间小鼠胸腺CD4+CD25+Foxp3-GITR+细胞及CD4+CD25+Foxp3+GITR+细胞占CD4+T细胞百分比无明显变化。与DMSO相比,DPN组的CD4+CD25+Fxop3+Treg细胞占CD4+T细胞百分比明显下降(P<0.05),表达GITR的Treg细胞下降(P<0.05),Treg细胞中GITR的平均荧光强度下降(P<0.05)。然而,与Oil组相比,E2组上述指标并无明显变化。结论:DPN单独激活ERβ后虽然不影响胸腺nTreg细胞的发育,却对CBA/J小鼠外周脾脏的Treg细胞起明显的抑制作用,提示ERβ单独激活可能促进自身免疫性疾病的发生、发展。
Objective:To study the effect of estrogen receptor beta(ERβ) on Treg cells in CBA/J mice. Methods: The per- centage of CD4 + CD25 + Foxp3 - GITR + T cells and CIM + CD25 + Foxp3 + GITR + T cells of C1M + T cells in the thymus of CBA/J mice, the percentage of CIM + CD25 + Foxp3 + Treg cells of CIM + T cells, the percentage of Treg cells expressing GITR and the GITR mean fluoresce intensity(MFI) in the spleen of CBA/J mice were detected by flow eytometry (FCM). Results: There was no significant difference in the percentage of CD4 + CD25 + Foxp3 - GITR + T cells and CIM + CD25 + Foxp3 + GITR + T cells of CD4 + T cells of the thy- mus in CBA/J mice neither between DPN group and DMSO group, nor between E2 group and Oil group. However, a significant de- crease of the percentage of CD4 + CD25 + Foxp3 + Treg cells of CIM +T cells( P 〈 0.05 ), the percentage of Treg cells expressing GITR ( P 〈 O. 05 ) and the GITR mean fluorescence intensity ( P 〈 O. 05 ) within the spleen was observed in DPN group compared with it' s control DMSO group. While there was no difference in the percentage of CIM + CD25 + Foxp3 + Treg cells of CIM + T cells, the percent- age of Treg cells expressing GITR and the GITR MFI between E2 group and it' s control Oil group. Conclusion : Although estrogen re- ceptor beta separate activation has no effect on the development of Treg cells in thymus, it does significantly suppress the Treg cells in the spleen, which suggests that estrogen receptor beta activated alone may promote the initiation and progression of autoimmune disea- ses.
