摘要 建立了高效液相色谱-蒸发光散射检测器(HPLC—ELSD)同时检测蚕蛹油中三种主要脂肪酸的方法。以Hanbon Lichrospher C18柱(5μm,250mm×4.6mm)为固定相,丙酮-水(2%乙酸)(v:v)为流动相,通过梯度洗脱实现目标产物的基线分离。针对各目标物建立工作曲线,亚麻酸、棕榈酸和油酸的线性范围分别为0.2~2.0mg/mL、0.1~2.0mg/mL和0.2-2.5mg/mL,R2〉0.99,检出限(S/N=3)均为0.05mg/mL,回收率97%-107%,相对标准偏差均小于2%。该方法快速、简便,可同时检测蚕蛹油中三种主要的脂肪酸。 A rapid and simple method was established for simultaneous determination of three main fatty acids in chrysalis oil based on high performance liquid chromatography-evaporative light-scattering detection (HPLC-ELSD). Chromatographic separation was carried out in gradient mode by using a Hanbon Lichrospher C18 column (5μm, 250mm ×46mm) with mobile phases of acetone-water (2% acetic acid ) (v/v). The fatty acids were in baseline separated, respectively. The calibration curves were plotted on the double logarithmic scales. The linear ranges of linolenic acid,palmitic acid and oieic acid were 0.2-2.0g/mL,0.1 -2.0mg/mL and 0.2- 2.5mg/mL, respectively. R2〉0.99,the limits of detection (S/N =3) were 0.05mg/mL,the recoveries were 97% 107% and the relative standard deviations were below 2%. The analytical method established in this work could be applied to accurate and rapid determination of three fatty acids in chrysalis oil.
