摘要
研究微波辅助提取,反相高效液相色谱测定花菜eevc含量的方法。花菜习浆后,以1%草酸为萃取溶剂,微波提取、净化后,采用恒流的方法进行液相色谱定量检测,以0.1%草酸溶液为流动相,流速1.0mL/min,采用C18反相色谱柱分离,在254nm波长下检测。结果表明,VC质量浓度在0.02-43.12mg/mL范围内线性良好,相关系数为0.9996,检测限0.02mg/kg,回收率在100.5%104.6%之间,相对标准偏差为1.20%。该方法能有效提取蔬菜中VC,测定结果稳定,重现性好,可以用于蔬菜中VC的检测。
The content of vitamin C in cauliflower was determined by microwave and HPLC. Vitamin C was extracted by microwave-assisted extraction of with 1% oxalic acid, then analyzed with HPLC by controlling the speed of mobile phase. Mobile phase was 0.1% oxalic acid with flow rate 1.0 mL/min; vitamin C was separated by C18 reversed column and detected by UV detector at 254 nm. It was showed that vitamin C in cauliflower could be separated. The linear ranges were 0.02-0.12 mg/mL, and the obtained correlation coefficient was 0.999 6. The recovery for the vitamin C ranged from 100.5%-104.6%, with the relative standard deviation (RSD) of 1.20%. The detection limits was 0.02 mg/kg. This method had good precision and high accuracy, thus can be applied to determine vitamin C in vegetables.
