清热化痰法对气道黏液高分泌大鼠气道炎症影响机制 被引量:3

摘要 目的:研究清热化痰法对气道黏液高分泌大鼠气道炎症的影响及机制。方法:将48只SD大鼠随机分为6组,每组8只,分别为空白组、模型组、低剂量止咳颗粒组、中剂量止咳颗粒组、高剂量止咳颗粒组、对照组,采取BALF细胞计数及分类观察炎症细胞、HE染色观察肺组织病理变化及ELISA检测血清NE等方法。结果:①细胞计数及分类:空白组、中、高剂量止咳颗粒组中性粒细胞、嗜酸性粒细胞、淋巴细胞较模型组、低剂量止咳颗粒组低(P<0.05或P<0.01)。②HE染色:正常大鼠气道黏液较少;模型组大鼠炎性细胞浸润及分泌物阻塞气道,肺泡壁多处断裂,部分肺泡融合成肺大泡;低剂量止咳颗粒组较模型组无明显变化;中剂量止咳颗粒组气管、支气管炎症细胞浸润减少;高剂量止咳颗粒组支气管管壁分泌物堵塞少,上皮细胞排列尚整齐;对照组肺泡壁断少,偶可见少许炎细胞。③血清NE:空白组、高剂量止咳颗粒组血清NE含量较模型组、低、中剂量组止咳颗粒组低(P<0.01)。结论:杏杷止咳颗粒能够通过清热化痰法减轻气道黏液高分泌大鼠气道炎症,其机制可能与血NE下降有关。 rats with high airway mucus secretion and its mechanism. Methods : 48 SD rats were randomly divided into 6 groups: the blank group, model group, low dose, middle dose and high dose of cough particle groups and the control group, 8 .in each group. The BALF cell count and classification were used to observe the inflammatory cells and HE dyeing for the observation of lung tissue pathology. ELISA method was used to detect serum NE, etc. Results : ( 1 ) Cell count and classification: In blank group and middle and high dose of cough particles groups, neutrophils, eosinophils and lymphocyte were lower than those of the model group ( P〈0.05 or P〈0.01 ). ( 2 ) HE stain : There was less airway mucus in normal rats. Model group rats blocked airway inflammatory cells infiltration and secretion with large area of alveolar walls fracture and part of the alveolar fusion into pulmonary bullae. Low doses of cough particles group compared with model group, there was no obvious change. Middel cough particle group's tracheal and bronchial inflammation cells infiltrations were reduced. High close cough particles group's bronchial wall secretion was less and the epithelial cell arrangement was neat. Control group's alveolar walls were broken a little, even some visible inflammation cells. ( 3 ) Serum NE : blank group and high dose of cough particles group's NE level were lower compared with those of the model group and low dose of cough particles group ( P〈0.01 ). Conclusion : Cough particles can reduce high airway mucus secretion and the mechanism may be associated with reducing blood NE.
