续骨活血汤对家兔骨折愈合影响实验研究

Study on Effect of Xugu Huoxue Decoction on Rabbit Fracture Healing

目的:观察续骨活血汤对家兔骨折愈合中血清骨钙素(BGP)、骨形态蛋白-2(BMP-2)表达和血液流变学的影响。方法:将108只家兔建立骨折模型,随机分为模型组、伤科接骨片组和续骨活血汤组,每组36只,各组均于造模术后第2天开始给药,2次/d,模型组予蒸馏水灌胃,伤科接骨片组予伤科接骨片200 mg/kg以混匀灌胃,续骨活血汤组予续骨活血汤浓缩口服液对人等效剂量的 2 倍灌胃。术后2周时随机抽取各组6只家兔,用自动血黏度测试仪检测血液流变学指标。于术后1、2、3、4、6周时随机抽取各组6只家兔,采用放射免疫法测定血清中骨钙素含量,再取材,进行免疫组织化学染色,观察骨折愈合不同时间点外骨膜、内骨膜和骨髓腔骨BMP-2的表达情况。结果:血液流变学及BGP、BMP-2的表达,续骨活血汤组、伤科接骨片组大部优于模型组,P<0.01或P<0.05;续骨活血汤组与伤科接骨片组比较,血液流变学大部时间点优于伤科接骨片,P<0.01或P<0.05;BGP和BMP-2的表达,部分时间点高于伤科接骨片组,P<0.01或P<0.05。结论:续骨活血汤对家兔骨折愈合有较好促进作用。

Objective ： To observe the influence of Xugu Huoxue Decoction on BGP and BMP-2 expressions as well as blood rheology of rabbits with fracture healing. Methods ： 108 rabbits were used to establish the fracture models and randomly divided into model group, Jiegu Tablet group and Xugu Huoxue Decoction group, 36 in each group. All groups were given drugs from the second day of modeling twice a day. The model group was given distilled water. Jiegu Tablet group was given Jiegu Tablet with the dose of 200 mg/kg. Xugu Huoxue Decoction group was given the double dose of human administration. 2 weeks after operation,6 rabbits of each group were taken out to test the blood rheology. 1,2,3,4, 6 weeks after operation,6 rabbits of each group were tested the BGP by radioimmnnoassay. And then by Immunohistochemistry method, BMP-2 expressions were observed. Results： BGP and BMP-2 expressions in Xugu Huoxue group and Jiegu Tablet group were superior to those of model group （ P〈0.01 or P〈0.05 ）. Xugu Huoxue Decoction group＇s blood rheology of most time was superior to those of Jiegu Tablet group （ P〈0.01 or P〈0.05 ）. BGP and BMP-2 expressions in part time were higher than those of Jiegu Tablet group （ P〈0.01 or P〈0.05 ）. Conclusion ： Xugu Huoxue Decoction has good improving effect of fracture healing of rabbits.
